However, if there are too many compliments, it can cause tissue damage and trigger an allergic reaction. C4A is an activation protein, which means it also activates the other complement proteins to increase in level [ 1 ]. C4a activates Neutrophils, which increases inflammation and free radicals [ 4 ]. You can request that your doctor test your C4A. Sometimes, a lab result may be in the reference range, but not actually be in the optimal range. These pathways are activated by infections, which include bacteria, fungi, viruses, and parasites. The body often detects infections by the toxins that they give off, so toxins may also increase C4a. One must realize, however, that C3A and C4A and general markers, and will never be able to diagnose Lyme disease specifically see images above.
One study found that C4a goes up after exercise in people with CFS [ 15 ]. A case study found that a CFS patient felt better after their C4a normalized [ 7 ].Forums New posts Search forums Google site search Popular content.
It was done because I had all the crazy fancy lyme tests and they were inconclusive. I could look up the paperwork and elaborate if needed He said it could be high because of lyme, as the tests could have been done when the lyme was mostly hiding InvertedTree Senior Member Nov 9, Messages Likes C4a Here's an article that discusses C4a.
Mine was high also. Thank you much, Inverted tree that cleared up some of my questions I think I have a few more but my brain is not in articulation mode. Messages 1, Likes Location twitpic.

C4a C4a is a measurement of a particular kind of complement. Complement is a part of the innate immune system. It identifies substances that the body doesn't want to let in. Complement can help to remove such foreign substances itself.
It also can alert other parts of the system to come help to remove the substance. Anaphylactic reactions such as ones that people get to peanuts, bee stings or latex are complement spikes. To my knowledge, the only thing that makes C4a rise is mycotoxins exposures poisons made by mold. This gene encodes the acidic form of complement factor 4, part of the classical activation pathway.
The protein is expressed as a single chain precursor which is proteolytically cleaved into a trimer of alpha, beta, and gamma chains prior to secretion. The trimer provides a surface for interaction between the antigen-antibody complex and other complement components. The alpha chain is cleaved to release C4 anaphylatoxin, an antimicrobial peptide and a mediator of local inflammation. Deficiency of this protein is associated with systemic lupus erythematosus and type I diabetes mellitus.
Varying haplotypes of this gene cluster exist, such that individuals may have 1, 2, or 3 copies of this gene. Two transcript variants encoding different isoforms have been found for this gene. Derived from proteolytic degradation of complement C4, C4a anaphylatoxin is a mediator of local inflammatory process. It induces the contraction of smooth muscle, increases vascular permeability and causes histamine release from mast cells and basophilic leukocytes.
Non-enzymatic component of C3 and C5 convertases and thus essential for the propagation of the classical complement pathway. Covalently binds to immunoglobulins and immune complexes and enhances the solubilization of immune aggregates and the clearance of IC through CR1 on erythrocytes.
C4A isotype is responsible for effective binding to form amide bonds with immune aggregates or protein antigens, while C4B isotype catalyzes the transacylation of the thioester carbonyl group to form ester bonds with carbohydrate antigens.
Systemic lupus erythematosus SLE : A chronic, relapsing, inflammatory, and often febrile multisystemic disorder of connective tissue, characterized principally by involvement of the skin, joints, kidneys and serosal membranes.
It is of unknown etiology, but is thought to represent a failure of the regulatory mechanisms of the autoimmune system. The disease is marked by a wide range of system dysfunctions, an elevated erythrocyte sedimentation rate, and the formation of LE cells in the blood or bone marrow. Complement component 4A deficiency C4AD : A rare defect of the complement classical pathway associated with the development of autoimmune disorders, mainly systemic lupus with or without associated glomerulonephritis.
Cytogenetic Location: 6p Molecular Location: base pairs 31, to 32, on chromosome 6 Homo sapiens Updated Annotation Release Complement tests are ordered by medical providers in order to determine the presence of an abnormality or a deficiency within the complement system. It may be used to look at pathways or components that could be leading to disease development or a deteriorating condition. One of the most common forms of complement system testing is the C4 complement blood test result.
To help with the diagnostic process of microbial infections that occur on a regular basis. To help monitor complex-related conditions or diseases such as vasculitis. A medical provider will order the C4 complement blood test when there is edema or inflammation of an unexplained nature. The complement system helps the immune system eliminate pathogens that may have invaded the body.
C4 levels are just one of the complement measures that are looked at. C3, CH50, and CH blood tests may also be ordered for a complete look at the immune system to determine at least a guess as to what may be happening medically. If someone has been to a medical provider several times within a short amount of time because of recurring bacterial infections, then a follow-up appointment for the C4 complement blood test is often a good idea.
Any time there are symptoms that could be tracked to an autoimmune disorder is also a good time to speak with a doctor. Inflammation that occurs for some reason without explanation may also trigger this blood test order.
People who have certain symptoms of lupus should immediately approach their medical provider for this test. This includes mouth ulcers, hair loss, a butterfly rash across the checks, and unusual joint pain or swelling. No preparation is usually necessary for this blood test to be completed. It can be drawn the same day that it is ordered. The C3 blood test is often ordered with the C4 complement blood test. All components from C1-C9 may be requested.
The C4 complement blood test can be abnormally high or abnormally low. Most people, however, will have a test result that comes back as normal. If the test results show that there is a C4 deficiency, then it can mean there could be a number of potential health concerns present.
The most common would be a bacterial infection. There may also be these additional health concerns based on an individualized medical history. When there is an increased level of C4 in the blood test results, then this is generally an indication that there is inflammation happening somewhere within the body.
This may be acute or chronic in nature. Ulcerative colitis may also cause increased complement levels. Having an increased or decreased C4 complement level cannot give a medical provider an idea of what is wrong with someone.
Men and women have different levels that are considered to be normal.Note: Turn around times on results are an estimate and are not guaranteed. Thank you so much for taking the time to leave a review : We really appreciate it - Michelle. I purchased Standard Wellness Test. Excellent and fast service. My results were ready on the next day after collecting the blood.
I thought levels of ferritin, iron, B12, B9 will be showed in this test as well. But no. Big disappointment for me. Unfortunately the Standard Wellness Test as outlined on Groupon does not show those levels. We hope to serve you again in the future - Nate. We are happy to hear about your experience. We are always honored to serve - Nate. The company did a great job getting me in and out with my appointment time plus the customer service was great.

The only diagnosis they gave him was he was all of a sudden at five years old autistic…although he has never shown any signs of being on the spectrum!
In a search to get him help with over twenty doctors, we finally saw an integrative doctor out in Connecticut who ran over thirteen vials of blood on him and sixteen on me. Enter C3a and C4a, the new kids on the block in the world of Lyme diagnosis and treatment. Complement proteins work with antibodies to destroy pathogens. They activate immunity through control of inflammation, phagocytosis ingestion of pathogens by white blood cells and cell death by lysis breaking of the cell membrane.
Both of these complement products may be increased in normal pregnancy and in certain types of vasculitis an inflammatory condition that destroys blood vessels.Arq backup linux
C4a levels are elevated in adult insulin dependent diabetes. Those who suspect that chronic fatigue syndrome CFS may actually be misdiagnosed Lyme disease may not be surprised to hear that C4a is also elevated in CFS patients. The normal range for the C4a is zero to In my chronically ill Lyme patients, I have seen C4a levels as high as about 26, However, most of my patients start with a pre-treatment level between and 12, As you can see by my test above my C4A test was over 16,!

If you would like your health care provider to order your C3a and C4a levels, it is extremely important that the tests be performed only at the National Jewish Medical and Research Center Laboratory in Denver. LabCorp uses two different send-out labs for the test and it is important to indicate that samples should be routed to National Jewish for most accurate results. To find the LabCorp drawing station nearest you, go to www. If your health care provider writes the ICD-9 diagnostic code of You may remember that white blood cells a.
White blood cells can be categorized as either granulocytes or mononuclear leukocytes. Mononuclear leukocytes are further sub-grouped into monocytes and lymphocytes. Lymphocytes, found in the blood, tissues, and lymphoid organs, attack antigens foreign proteins in different ways. The main lymphocyte sub-types are B-cells, T-cells and natural killer NK cells. B-cells make antibodies that are stimulated by infection or vaccination. T-cells and NK cells, on the other hand, are the cellular aggressors in the immune system and are our main focus in the discussion that follows.
NK cells have their own specific surface markers. The predominant marker is CD I hope this post helps you get to the bottom of your mysterious symptoms and to get some answers before it is too late. I ended up getting answers with an integrative doctor over a western medicine doctor because after of years of doing the basic Lyme testing and western blots that all showed negative Lyme scores. The reason for that is a normal lyme test shows your bodies antibody response to fighting a lyme infection, the only problem was my body was never fighting the infection, so the infection had killed my fighter cells so it would never show up on basic lyme testing.
So if you have done the basic testing and your doctor is still saying that your symptoms are in your head and you know they are not, I highy recommend getting these above test that are covered under most insurances! Thanks for posting this, we are in the thick of it with a CD57 of 20 and the UK not really hearing it at all, this is a really useful and clear explanation.

I know there have been discussions on this board about the C4a level but I haven't been able to find anyone who was able to lower their C4a level. I know it's either Lyme, Mold or some other infection that is causing the elevated level and it's an inflamatory response that totally messing my body up!
I would love to hear from people who have dealt with this and what results they got? How did they go about identifying which infection they had or if it was a Mold issue? Thanks a bunch. Lono83 New Member. Here are some of the things that I've heard people have had success with re: lowering C4a - mold avoidance, antibiotic treatments, Procrit I believe only Dr.
Shoemaker uses this. My opinion is that there's not a great way to know whether the high C4a is being caused by an infection things like AIDS and Lyme have been shown to increase C4a or mold exposure.
I think all you can do is try to treat a number of different things and see if there's improvement. You should also get your TGF-Beta1 tested. If that's high then you'll want to pay attention to it and try and get it lower.Indiana snitch watch
If these are normal, and if you don't have any of the HLA genotypes that Shoemaker pays attention to, then from patient reports I've heard, it seems easier to get improvements. My C4a is quite high, over 15k, and I haven't had luck getting it down over the last year.
I probably have some mold exposure at my job, but it's not really feasible at the moment to get different employment. I also might have a persistent infection, but there's nothing I really trust that can prove this definitely. A couple other things you can do right now -- chances are your sleep is messed up. Do what you have to do to get decent sleep.
I have to take a sleeping pill every night which I hate but if I didn't my fatigue would be much worse.
